How this all works in theory:
- I spend 2 weeks and use liters of media to generate a sufficient amount of each sample.
- I submit my precious samples to the core facility (CF).
- CF takes the samples through a 3-step process.
- CF sends me the resulting data.
How this works in practice:
- I spend 2 weeks and use liters of media to generate a sufficient amount of each sample.
- I submit my precious samples to the core facility (CF).
- CF does nothing with my samples for several weeks.
- CF tells me one of their reagents went bad, and 2 out of 4 of my samples were destroyed in Step 1.
- I start remaking the 2 lost samples.
- CF tells me an error occurred with one of the remaining samples during Step 2, and although they were able to do Step 3 with it, the resulting dataset is incomplete.
- I start remaking the third lost sample.
- No news on my last surviving sample from the first submission.
- I hear from someone in another lab that CF has a huge backlog in their pipeline, which I guess explains the silence.
- CF finally sends me data from the last sample. A 25% success rate...woohoo!
- I submit the 3 newly-prepped replacement samples to CF.
- CF does nothing with my samples for 2 weeks.
- CF tells me that 2 of my 3 replacement samples have insufficient material by their quantitation method. They were fine by my quantitation method, which was the same method I used for the first submission, when there was no discrepancy between my measurements and theirs. CF refuses to proceed with these two samples until more material is submitted.
- I start making supplemental samples for the 2 replacement samples.
- CF says the third replacement sample is borderline in quantity, and that they can proceed but will have to modify their normal protocol to make it work.
- I tell CF that since I'm already making more of the first 2 samples, I'll just make more of the third one too since I'd rather they use their normal protocol.
- CF tells me that oops, they've already begun Step 1 with the third sample so no more material can be added at this point.

I'm pretty sure there is a national Core Facility conference where they exchange ideas on fucking with us. There are probably talks and symposia with titles like "Breaking the will of junior faculty" and "It's not my problem, it's yours". They probably have to hold it in Vegas because the attendance is so high.

I've given up on sending my precious samples to our core facility and have started using a commercial set-up nearby. Yes, it costs more money but does wonders for my stress levels!
Academic core facilites have a tendency towards medicority, mostly because--unlike in commercial outfits--no one ever gets fired for incompetence.
Good grief! CF is a special kind of hell I wish I could avoid. I don't understand how two sets of people can get such varying amounts of data, where set 1 = people who actually generate the sample and set 2 = core facility people.
Time to find another -- read: commercial -- facility which will do said protocol with a minimum of problems. In the long run, it may even turn out to be more cost effective.

Tig and Tom--Yeah, I've definitely thought about it, but we get a pretty big discount here along with a few other "special considerations". And of course, there are politics to consider....
CPP--Not to mention the fact that the faculty member who is in charge of the facility, and therefore supposedly responsible for its operations, is tenured. Can't fire the employees, can't fire the boss.
MXX--Ugh...I'd avoid it too if I could! I don't know why the measurements are so different. Sometime next week I may call around the department and see if anyone else has had this particular problem.
To hell with the politics. If they're offering crap service, fuck 'em. Politics are all well and good up until the point that they prohibit you from advancing your career ... and shit service cuts down your ability to rapidly produce data, which cuts down on your ability to rapidly produce publications.
Besides, what good is a discount if they keep fucking up your experiments, causing you to spend even more money on gallons of media? If you do the math, you may find out it's a total wash and the commercial company will give you a better rate ... besides, if you explain the commercial company that you'd like to try their service, but they're somewhat cost prohibitive ... they may cut you a discounted rate.
Sorry to hear about Core Facilty hell. Very frustrating to be so dependent on others. As Thomas Joseph said, time to start shopping around?

Bean-mom--Yes, definitely frustrating to be dependent. We don't have the very expensive machine required to do this procedure, so it's not even like I can just learn to do it myself. Thanks for the sympathy!

Arlenna--Actually, no, but our proteomics facility has an ever worse reputation than the facility I'm complaining about, so it definitely could've been!
